This variant lies in the MCCC1 gene (transcript NM_020166.5) at coding-DNA position 215, where G is replaced by C; at the protein level this means replaces glycine at residue 72 with alanine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 72 of the MCCC1 protein (p.Gly72Ala). This variant is present in population databases (rs761521333, gnomAD 0.003%). This missense change has been observed in individual(s) with 3-methylcrotonyl-CoA carboxylase (3MCC) deficiency (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1399715).
