NM_201384.3(PLEC):c.10918T>G (p.Tyr3640Asp) was classified as Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2Q; Epidermolysis bullosa simplex, Ogna type; Epidermolysis bullosa simplex with nail dystrophy; Epidermolysis bullosa simplex 5C, with pyloric atresia; Epidermolysis bullosa simplex 5B, with muscular dystrophy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with PLEC-related conditions. This sequence change replaces tyrosine with aspartic acid at codon 3667 of the PLEC protein (p.Tyr3667Asp). The tyrosine residue is moderately conserved and there is a large physicochemical difference between tyrosine and aspartic acid. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
