Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.182G>A (p.Cys61Tyr), citing Ambry Variant Classification Scheme 2023: The p.C61Y variant (also known as c.182G>A), located in coding exon 1 of the TMEM127 gene, results from a G to A substitution at nucleotide position 182. The cysteine at codon 61 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
