NM_001378454.1(ALMS1):c.2714C>G (p.Ser905Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S906C variant (also known as c.2717C>G), located in coding exon 8 of the ALMS1 gene, results from a C to G substitution at nucleotide position 2717. The serine at codon 906 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
