Uncertain significance for Atrial septal defect 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004387.4(NKX2-5):c.847C>G (p.Pro283Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NKX2-5 gene (transcript NM_004387.4) at coding-DNA position 847, where C is replaced by G; at the protein level this means replaces proline at residue 283 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline with alanine at codon 283 of the NKX2-5 protein (p.Pro283Ala). The proline residue is moderately conserved and there is a small physicochemical difference between proline and alanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The alanine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with NKX2-5-related conditions. This variant is not present in population databases (ExAC no frequency).
