Uncertain significance for Deafness-lymphedema-leukemia syndrome; Monocytopenia with susceptibility to infections — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032638.5(GATA2):c.368A>G (p.Lys123Arg), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces lysine with arginine at codon 123 of the GATA2 protein (p.Lys123Arg). The lysine residue is moderately conserved and there is a small physicochemical difference between lysine and arginine. This variant has not been reported in the literature in individuals affected with GATA2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
