Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004525.3(LRP2):c.8219C>T (p.Thr2740Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with isoleucine at codon 2740 of the LRP2 protein (p.Thr2740Ile). The threonine residue is highly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with LRP2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
