This variant lies in the GMPPB gene (transcript NM_021971.4) at coding-DNA position 854, where G is replaced by T; at the protein level this means replaces cysteine at residue 285 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces cysteine with phenylalanine at codon 285 of the GMPPB protein (p.Cys285Phe). The cysteine residue is weakly conserved and there is a large physicochemical difference between cysteine and phenylalanine. This variant is not present in population databases (ExAC no frequency).
