Pathogenic for Early-infantile DEE — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006279.5(ST3GAL3):c.302+1del, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Phe102Serfs*34) in the ST3GAL3 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in ST3GAL3 are known to be pathogenic (PMID: 31584066). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with ST3GAL3-related conditions. This variant is also known as c.302+1del. This variant is not present in population databases (ExAC no frequency).
