Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.2110A>G (p.Lys704Glu), citing Ambry Variant Classification Scheme 2023: The p.K704E variant (also known as c.2110A>G), located in coding exon 12 of the DICER1 gene, results from an A to G substitution at nucleotide position 2110. The lysine at codon 704 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
