Uncertain significance for Hereditary spastic paraplegia 28 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001160148.2(DDHD1):c.700G>T (p.Ala234Ser), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 234 of the DDHD1 protein (p.Ala234Ser). This variant has not been reported in the literature in individuals affected with DDHD1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
