Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005502.4(ABCA1):c.1973G>T (p.Gly658Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with valine at codon 658 of the ABCA1 protein (p.Gly658Val). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and valine. This variant is present in population databases (rs758163019, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with ABCA1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ABCA1 protein function. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
