Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001277115.2(DNAH11):c.10737T>G (p.Phe3579Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine with leucine at codon 3579 of the DNAH11 protein (p.Phe3579Leu). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and leucine. This variant is present in population databases (rs369261194, ExAC 0.002%). This variant has not been reported in the literature in individuals with DNAH11-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
