Pathogenic for Familial adenomatous polyposis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.9:g.(?_112102013)_(112103097_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with APC-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 3-4 of the APC gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in APC are known to be pathogenic (PMID: 17963004, 20685668). For these reasons, this variant has been classified as Pathogenic.
