NM_172232.4(ABCA5):c.4320+1G>C was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.4320+1G>C intronic alteration consists of a G to C substitution one nucleotide after exon 32 (coding exon 32) of the ABCA5 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of ABCA5 has not been established as a mechanism of disease. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
