Uncertain significance for Short-rib thoracic dysplasia 14 with polydactyly; Joubert syndrome 23 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001329943.3(KIAA0586):c.2938G>C (p.Asp980His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the KIAA0586 gene (transcript NM_001329943.3) at coding-DNA position 2938, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 980 with histidine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 1033 of the KIAA0586 protein (p.Asp1033His). This variant is present in population databases (rs754818478, gnomAD 0.001%). This variant has not been reported in the literature in individuals affected with KIAA0586-related conditions. ClinVar contains an entry for this variant (Variation ID: 1395462). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on KIAA0586 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
