This variant lies in the KIF1A gene (transcript NM_001244008.2) at coding-DNA position 785 through the canonical splice donor site of the intron immediately after coding-DNA position 798, deleting this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. ClinVar contains an entry for this variant (Variation ID: 1395436). This variant has been observed in individual(s) with clinical features of KIF1A-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This variant, c.784_798del, results in the deletion of 5 amino acid(s) of the KIF1A protein (p.Gly262_Lys266del), but otherwise preserves the integrity of the reading frame.
