NM_015450.3(POT1):c.280C>G (p.Gln94Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q94E variant (also known as c.280C>G), located in coding exon 4 of the POT1 gene, results from a C to G substitution at nucleotide position 280. The glutamine at codon 94 is replaced by glutamic acid, an amino acid with highly similar properties. This alteration was identified in an individual diagnosed with melanoma (Robles-Espinoza CD et al. Nat Genet, 2014 May;46:478-481). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
