NM_203446.3(SYNJ1):c.614C>T (p.Ser205Leu) was classified as Uncertain significance for Developmental and epileptic encephalopathy, 53; Early-onset Parkinson disease 20 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with leucine at codon 244 of the SYNJ1 protein (p.Ser244Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs747414228, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with SYNJ1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
