This variant lies in the IHH gene (transcript NM_002181.4) at coding-DNA position 77, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 26 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Trp26*) in the IHH gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in IHH cause disease. This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This premature translational stop signal has been observed in individual(s) with brachydactyly (Invitae). In at least one individual the variant was observed to be de novo.
