Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.1120C>G (p.His374Asp), citing Ambry Variant Classification Scheme 2023: The p.H374D variant (also known as c.1120C>G), located in coding exon 5 of the BLM gene, results from a C to G substitution at nucleotide position 1120. The histidine at codon 374 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
