Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CACNA1H protein function. This variant has not been reported in the literature in individuals with CACNA1H-related conditions. This variant is present in population databases (rs771028729, ExAC 0.002%). This sequence change replaces glycine with arginine at codon 2298 of the CACNA1H protein (p.Gly2298Arg). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and arginine.
