This variant lies in the PKP2 gene (transcript NM_001005242.3) at coding-DNA position 464, duplicating one base; at the protein level this means shifts the reading frame starting at serine residue 155, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: ClinVar contains an entry for this variant (Variation ID: 1394074). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with PKP2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ser155Argfs*61) in the PKP2 gene. It is expected to result in an absent or disrupted protein product.
