Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.458C>A (p.Pro153His), citing Ambry Variant Classification Scheme 2023: The c.458C>A (p.P153H) alteration is located in exon 3 (coding exon 3) of the COL5A1 gene. This alteration results from a C to A substitution at nucleotide position 458, causing the proline (P) at amino acid position 153 to be replaced by a histidine (H). Based on data from gnomAD, the A allele has an overall frequency of <0.001% (1/247646) total alleles studied. The highest observed frequency was 0.006% (1/16166) of African alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
