NM_000314.8(PTEN):c.801+2T>C was classified as Pathogenic for PTEN hamartoma tumor syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PTEN gene (transcript NM_000314.8) at the canonical splice donor site of the intron immediately after coding-DNA position 801, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site results in skipping of exon 7 or use of a cryptic donor site and introduces a premature termination codon (PMID: 9600246, 11071384). The resulting mRNA is expected to undergo nonsense-mediated decay. Disruption of this splice site has been observed in individuals with PTEN-related conditions (PMID: 9600246, 10234502, 11071384). This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 7 of the PTEN gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product.
