Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_201590.3(CACNB2):c.43A>T (p.Ile15Phe), citing Ambry Variant Classification Scheme 2023: The p.I15F variant (also known as c.43A>T), located in coding exon 1 of the CACNB2 gene, results from an A to T substitution at nucleotide position 43. The isoleucine at codon 15 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
