Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_94514404)_(94514533_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 18 of the ABCA4 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individual(s) with Stargardt disease (PMID: 12754711). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant. Experimental studies have shown that a similar copy number variant affects ABCA4 function (PMID: 12754711). For these reasons, this variant has been classified as Pathogenic.
