Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172240.3(POC1B):c.1070C>T (p.Ser357Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with phenylalanine at codon 357 of the POC1B protein (p.Ser357Phe). The serine residue is moderately conserved and there is a large physicochemical difference between serine and phenylalanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with POC1B-related conditions. This variant is not present in population databases (ExAC no frequency).
