Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000077.5(CDKN2A):c.62C>A (p.Ala21Asp), citing Ambry Variant Classification Scheme 2023: The p.A21D variant (also known as c.62C>A), located in coding exon 1 of the CDKN2A gene, results from a C to A substitution at nucleotide position 62. The alanine at codon 21 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
