NM_001148.6(ANK2):c.10264G>A (p.Glu3422Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E3422K variant (also known as c.10264G>A), located in coding exon 38 of the ANK2 gene, results from a G to A substitution at nucleotide position 10264. The glutamic acid at codon 3422 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
