Uncertain significance for Congenital disorder of glycosylation, type IAA — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138459.5(NUS1):c.674C>T (p.Thr225Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 225 of the NUS1 protein (p.Thr225Met). This variant is present in population databases (rs760187558, gnomAD 0.008%). This missense change has been observed in individual(s) with clinical features of NUS1-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 1391842). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The methionine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
