Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152906.7(TANGO2):c.605+4A>C, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TANGO2 gene (transcript NM_152906.7) at 4 bases into the intron immediately after coding-DNA position 605, where A is replaced by C. Submitter rationale: This sequence change falls in intron 7 of the TANGO2 gene. It does not directly change the encoded amino acid sequence of the TANGO2 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (no rsID available, gnomAD 0.004%). This variant has been observed in individual(s) with clinical features of TANGO2-related conditions (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1391806). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
