Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4424C>T (p.Thr1475Ile), citing Ambry Variant Classification Scheme 2023: The p.T1475I variant (also known as c.4424C>T), located in coding exon 22 of the DICER1 gene, results from a C to T substitution at nucleotide position 4424. The threonine at codon 1475 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
