Uncertain significance for Alagille syndrome due to a JAG1 point mutation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000214.3(JAG1):c.757T>C (p.Cys253Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with arginine at codon 253 of the JAG1 protein (p.Cys253Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with Alagille syndrome (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
