This variant lies in the SDHD gene (transcript NM_003002.4) at coding-DNA position 124, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 42 with lysine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with lysine at codon 42 of the SDHD protein (p.Glu42Lys). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs756995021, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with SDHD-related conditions.
