Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001735.3(C5):c.4321G>T (p.Ala1441Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1441 of the C5 protein (p.Ala1441Ser). This variant is present in population databases (rs143886761, gnomAD 0.05%). This variant has not been reported in the literature in individuals affected with C5-related conditions. ClinVar contains an entry for this variant (Variation ID: 1391043). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
