Uncertain significance for Osteogenesis imperfecta type I; Ehlers-Danlos syndrome, classic type, 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000089.4(COL1A2):c.1448C>A (p.Ala483Asp), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the COL1A2 gene (transcript NM_000089.4) at coding-DNA position 1448, where C is replaced by A; at the protein level this means replaces alanine at residue 483 with aspartic acid — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 483 of the COL1A2 protein (p.Ala483Asp). This variant is present in population databases (rs414408, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with COL1A2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1390970). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on COL1A2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
