Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000143.4(FH):c.95C>T (p.Ala32Val), citing Ambry Variant Classification Scheme 2023: The p.A32V variant (also known as c.95C>T), located in coding exon 1 of the FH gene, results from a C to T substitution at nucleotide position 95. The alanine at codon 32 is replaced by valine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
