Uncertain significance for T-B+ severe combined immunodeficiency due to JAK3 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000215.4(JAK3):c.1997TCA[1] (p.Ile667del), citing Invitae Variant Classification Sherloc (09022015): This variant, c.2000_2002del, results in the deletion of 1 amino acid(s) of the JAK3 protein (p.Ile667del), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with severe combined immunodeficiency (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
