Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016004.5(IFT52):c.569G>C (p.Cys190Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with serine at codon 190 of the IFT52 protein (p.Cys190Ser). The cysteine residue is moderately conserved and there is a moderate physicochemical difference between cysteine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with IFT52-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
