This variant lies in the TXNRD2 gene (transcript NM_006440.5) at coding-DNA position 430, where C is replaced by T; at the protein level this means replaces arginine at residue 144 with cysteine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
