This frequency is not significantly higher than estimated for disease-causing variants in KAT6B, allowing no conclusion about variant significance. However, the observation of 22 heterozygous controls is not consistent with the onset and severity of KAT6B-related conditions. To our knowledge, no occurrence of c.3718G>A in individuals affected with KAT6B-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
