This variant lies in the DYRK1A gene (transcript NM_001347721.2) at coding-DNA position 1699, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 567 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of DYRK1A-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln576*) in the DYRK1A gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 188 amino acid(s) of the DYRK1A protein.
