Pathogenic for Arginine:glycine amidinotransferase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001482.3(GATM):c.724_725del (p.Gly242fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gly242Lysfs*6) in the GATM gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GATM are known to be pathogenic (PMID: 11555793). This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with GATM-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
