Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006941.4(SOX10):c.1086del (p.Gln364fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SOX10 gene (transcript NM_006941.4) at coding-DNA position 1086, deleting one base; at the protein level this means shifts the reading frame starting at glutamine residue 364, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant is also known as c.1083delG. This frameshift has been observed in individual(s) with Waardenburg syndrome 2 (PMID: 27759048, 29407415). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change results in a frameshift in the SOX10 gene (p.Gln364Argfs*138). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 103 amino acid(s) of the SOX10 protein and extend the protein by 34 additional amino acid residues.
