Uncertain significance for Sulfite oxidase deficiency due to molybdenum cofactor deficiency type A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001358530.2(MOCS1):c.1894dup (p.Asp632fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the MOCS1 gene (p.Asp632Glyfs*43). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 5 amino acid(s) of the MOCS1 protein and extend the protein by 37 additional amino acid residues. This variant is present in population databases (rs747994218, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with MOCS1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
