NM_000023.4(SGCA):c.199C>G (p.Gln67Glu) was classified as Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2D by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 67 of the SGCA protein (p.Gln67Glu). This variant is present in population databases (rs753180048, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with SGCA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
