Pathogenic for Familial hemophagocytic lymphohistiocytosis 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006949.4(STXBP2):c.982C>T (p.Gln328Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the STXBP2 gene (transcript NM_006949.4) at coding-DNA position 982, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 328 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with STXBP2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Gln328*) in the STXBP2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in STXBP2 are known to be pathogenic (PMID: 19804848, 22451424).
