NM_198525.3(KIF7):c.1443+5C>T was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015. This variant lies in the KIF7 gene (transcript NM_198525.3) at 5 bases into the intron immediately after coding-DNA position 1443, where C is replaced by T. Submitter rationale: DNA sequence analysis of the KIF7 gene demonstrated a sequence change in intron 5, c.1443+5C>T. This change does not appear to have been previously described in individuals with KIF7-related disorders. This sequence change has been described in the gnomAD database with a frequency of 0.16% in the African/African American subpopulation and 0.02% in the overall population (dbSNP rs778312422). In-silico splice prediction programs provide inconclusive results for this sequence change. The functional significance of this sequence change is not known at present.
