Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001205293.3(CACNA1E):c.5045C>A (p.Thr1682Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with asparagine at codon 1682 of the CACNA1E protein (p.Thr1682Asn). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and asparagine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CACNA1E protein function. This variant has not been reported in the literature in individuals affected with CACNA1E-related conditions. This variant is present in population databases (rs775398717, ExAC 0.009%).
